Curiously in Destiny 1 and 2. Signs around the tower and the New Tower are typically placed in three languages. The first being English, the next Spanish or French, finally Chinese. This suggests that the primary ethic groups of The Last City are of English, Spanish, and Chinese heritage.

The population of the Last City has been in debate, apparently according to in game sources the last census taken found the city population to be around 675,000(citation needed). Of course, this was before the events of the Red War.
